Maybe not really making her a villain, but her actions have her sliding down the anti-hero scale so fast that Twilight starts having second thoughts whether they're doing the right thing.

I think it's somewhat possible. Somehow Celestia forgets about friendship (or if the show becomes even more focused on the world and characters than lessons in friendship, becomes jaded with her kingdom,) then she may become a Sun version of Nightmare Moon. I think the conflict of fighting The Princess and your mentor is within the scope of what the show would produce. As long as she's able to be turned back to normal, then I think it could happen. They'll write it to be much better than that though.
