The Mmmm lacked a solid structure, which was very obvious when moved. (Bear in mind that Mac's probably used to pulling or carrying things that are more stable i.e. carts, plows, etc.)

The reason Big Mac has trouble with the cake is because it's UNBALANCED, it's basicallly 3 cakes that is barely connected together. It's like trying to carry a stack of books, in which the problem isn't trying to carry them it's trying to keep them in balance so they won't topple over.

Don't make me link how everypony had trouble holding Twilight's weight when she was falling in the pilot, or RD carrying four other ponies at top speed. Lesson: ponies can carry as much as the plot demands.

There certainly was something strange about this cake, though.

Cakes can get pretty heavy when they get that big and when they're that tiered and when they're heavily decorated (has anyone seen one of those cake decorating contests on the Food Network once for fun when there was absolutely nothing else--not even ponies--on television? Those cakes require more than one person to even transfer it from the table to a dolly!); in theory, that cake could be as heavy as Tom. In addition, he had to struggle to keep it balanced like some others above me mentioned. Those feeble columns were hardly supporting it in the first place.

There's also the fact, that many others have pointed out before, that he's more of a puller than a lifter. Sure he has lifted before such as barrels of cider and buckets of towered apples, but most of that hauling power lies in his legs. Recall H&HD when he hit that tree with only one hoof and a plethora of apples fell and then later pulled Berry Punch's house (ignoring that he was on some sort of love fueled adrenaline).

In all seriousness, the Cakes should have just asked Twilight to teleport it into the train in the first place. And why are we arguing about a children's cartoon? :P But what do I know? I'm just a nameless Anon who'll get ripped apart by the account holding members of the site.

...why is there a thumb fitting on the oven mit hanging on the wall behind Big Mac?

If anything, it's more likely he was trying to be extra careful so that the cake doesn't get ruined. No one would want anything like that to get destroyed after all. Especially not while transporting it.
