I will explain the difference between one who has shipping goggles on for two particular characters by using myself as an example.
I see a romance transcending time and race, built from the moment these two met eyes for the first time, potentially facing perils to separate them but to otherwise spend their final days together content and no less in love than the day they wed. Normal people see a dragon and a pony cutely hugging. |

Its debatable whether they see each other as brother and sister but to the very least they are childhood friends. :) |

I think Twilight put it best when she said Spike was someone she could never replace, leaving it open what their exact relation is. |

^ I think its more her being a responsible older sister. I couldn't ship these two. It'd be a little too odd. Been raised since birth and probably having had a few woes and joys together. Twilight never had any friend but Spike until coming to Ponyville after all so he was the only person who could be there for her.

We'll probably find out in the coming up episode when Spike goes on his quest. Personally, I see them as being more than just friends, but not like lovers or a mother/son relationship. A close brother/sister relationship. Not all of such relationships get along well (like me and my sister, we're at each other's throats a lot of the time), but plenty do, and this is one of them. |

I always got the feeling that Spike and Twilight would always be friends. The sort of friends you complain about your relationships too.
I actually don't see them as brother and sister. Twilight isn't too much older than he is and her parents probably raised her until she was responsible enough. That being said, she is still quite a bit more mature. |
