Probably the same size as normal paper. Assuming the pony in question was writing with utensils ala Twilight's quill, each letter would probably fit in the same space as a human capital B.

At the same time there was probably a period of time in which ponies could only fit one letter onto a piece of paper, which probably means 90% of Equestria was deforested to supply enough parchment.

The Pegasus probably would be able to write using their feather similar to fingers. Unicorns could magically use a stick. This alphabet would probably be an ancient form of Earth Pony

And they would of course probably develop a sort of stamp that they could hold in their mouth or something consisting of a single hoofprint, making the letters by rotating it with their tongue.

Equestria clearly has two alphabets.
Spike's ignorance of words in the first letter doesn't seem to be based on difficulty, but simply not knowing what they mean. He, Twilight, and Celestia are some of the only ones who can read Ancient Equestrian, while everyone converses in the modern tongue. He didn't know how to translate "brink", as opposed to being unable to spell it.

^ The lines in "BB" and "BA" would not be contiguous - like "m" and "rn."
